Kaine was the first clone of Spider-Man created by the Jackal. He was suffering from clone degeneration and became an assassin and spent years tormenting his clone brother, Ben Reilly. He has since taken up the mantle his now dead brother as the new Scarlet Spider.
The Jackal is an enemy of Spider-Man who played a big role in both of the Clone Sagas. He has since seemingly returned to infest Manhattan with spider-powers for his own sinister ends, organizing test subjects into a spider-powered army.
Mary Jane Watson is an actress and a major love interest of Peter Parker aka Spider-Man. They have had a long and dramatic on-and-off-again relationship. She's one of the few people who know who he is and still supports him as a close friend and confidante.

He was once the chief editor and publisher of the Daily Bugle, a New York newspaper, and the mayor of New York before his resignation amidst scandal resulting from Superior Spider Man and the Goblin Nation. He now is a host of a show on the F.A.C.T. News Channel

Jason Philip Macendale, former agent of CIA, turned from the first Jack O'Lantern to the second major Hobgoblin and once the host of Demogoblin. He was rebuilt as a cyborg and remained Hobgoblin until the first Hobgoblin who was thought dead returned and killed him.
Spidercide is a clone of Spider-Man created by the Jackal (who has created the majority of the Spider-Man clones) who made Spidercide believe he was the true Peter Parker. Spidercide's insistence that he was the true Peter Parker led him down a road of violence and villainy.
