What does the title mean... well in this case, its an homage to the movie Best of the Best (late 80's Tae Kwan Do film). These characters are the best fighters in their respective companies. Yes, some might use weapons, but they also have proven they don't need weapons, gadgets or powers to win a fight.
The list is limited to modern era fighters though (no future characters or immortals here, long lived is ok though) and it is currently in no particular order.

I rank Cassie as #2 due to the fact that (this may be an oversight on my part) but I've never seen her beat Batman in a fight. She is great, there is no doubt about that, however Batman is the bar that all others are measured by.
Ranked 3rd as she can't beat her daughter and Batman manages to consistently best her.
This man has trained many of the DCU's best fighters. I actually place him tied with Lady Shiva as the only reason Dragon didn't kill Shiva the last time they fought was interference and a wish to deprive Neron of Shiva's soul.
Wolverine is the best there is at what he does. He is a master of various martial arts and sword styles. Not someone to mess with (looking at you X).
The Clone of Wolverine has been trained to kill, plain and simple. Mix this with the trigger scent and you'd best not be near her when she's in mindless kill mode.
The guy who thinks he is the best there is at killing. He finally met his match with Wolverine. He can't stand that.
Connor Hawke is the son of Oliver Queen, the original Green Arrow. In addition to Archery he is a upper level Martial Artist that has survived an encounter with Lady Shiva, not something many can say.
Daughter of the original Black Canary, Dinah is a dedicated martial artist that does not need to rely on her Canary Cry to help her against many of her opponents.
Not a damsel in distress thats for sure.
Many rank him above Connor but the two have not faced each other in unarmed combat. He has bested Oli but at the time Oli wasn't not known for his martial prowess.
There is not doubt that he is a real threat to those he faces.
