The Killing Joke is a retelling of the Book of Job casting Batman as God, Joker as Satan and Jim as Job.

In the Old Testament story, Satan makes a bet with God that if he torments one of His most devout followers enough, he'll renounce his faith, trying to prove that God is only loved and followed because of the riches He provides. So Satan goes down and just completely destroys Job's life, kills his family, destroys his house, strikes him with disease, everything he can think of. Job has a long argument with God, but then ultimately refuses to renounce Him, proving Satan wrong.

This is essentially what happens in The Killing Joke, and it feeds into the meta-narrative people like to point out about superheroes not caring about the people who get hurt in their battles. It seems like a very Alan Moore thing to do, even if it wasn't his intention.
