Round 1: Zeref will win. EMS Madara lacks the abiliity to kill an immortal.

Round 2: Madara will win. Even if Zeref could kill immortals, the Impure world Resurrection will keeps madara from dying permanently. Madara on the other hand has access with the abilities of the Rinnegan. Madara could use the King of Hell to separate Zeref's soul from his body.

Round 3 and 4 goes to madara as well. Even if zeref's ability managed to kill madara, Madara's Transcription Seal: Izanagi will activate and he will only repeat his action from Round 2.

Zeref wins 1st round, Madara wins the rest.

EMS Madara has also the power of Kyuubi Susanoo and we didn't see Zeref surviving complete vaporization caused by dozens of Bijuudama Blades storming on his head. Also basing on Zeref claming another punch from Igneel amped Natsu would have probably killed him.

After all even Ultear was able to KOing Zeref since Tenro Island.
