Optimus is far more skilled than Thanos. Prime can take on his equal with 2 other veteran decepticons and slaughters 16 Decepticon veterans in TRF 3 OPTIMUSSSSS Scene (Yes Sam).
Realistically Thor would have cut off Thanos's head in that three man fight in endgame, if not for Plot Physics. The only reason Thanos can even fight is because he hindsight his opponents moves before fighting them. So he knows Cap's moves, Stark's moves, Thor's moves, Hulk etc. Otherwise he's dead. That is why Stormbreaker nearly killed him in IW but Thanos foresaw in Endgame that he needed to dodge it rather than overpower it.
Stark in IW cuts Thanos with Superfists and overpowers Thanos's IG fist with his leg. Spidey holds back Thanos's unarmed IG fist with enough force to stall Thanos Badly. Spidey can hold a helicopter in Homecoming and a small clocktower together in Far From Home. Drax with kitchen knives can make Thanos Grunt when knee kicked, Quill stumbles Thanos with an Electromagnetic grenade and Nebulla slams a plane right into Thanos, stumbling him. Thanos is only boosted by the stones to make him survive a beating or without IG, he would be dead. Which is why he defeated the Hulk
Prime's energon Blades can easily Behead Thanos plus Sword of Judgement.

The IG operates in One Universe only and that is the MCU, it doesn’t work anywhere where Thanos wants to. He can’t snap Optimus if the being is not from the MCU, let alone from a different franchise. The comics state this and it is a Universal fact of the cosmic hierarchy of the MCU. The reality Stone would be useless as it doesn’t recognize Prime’s existence under the MCU so practically no impact same goes for soul stone, time stone - only for MCU time manipulation and not elsewhere etc.
The only stone that could work is the Power Stone which is Thanos’s opportunity but Prime has tanked Universal+ attacks being Quintessa and AllSpark Megatron (hurt him badly). Can fly far into cosmos and survive the total impact from Earth onto Cybertron which is light years away. In G1 it is 100 Million Light years away. Megatron can fly from Earth to Saturn in a few minutes - hours which is 746 Million Miles away. The TRF can dodge missiles point blank to their face, yes Ironhide you!
Unless Thanos has high heat rounds or “Sabot Rounds”, Thanos struggles to hurt Prime. You misunderstand the TRF Franchises. The TRF are weak to high heat rounds and that is why F-22 Rockets can even hurt Transformers. Otherwise The TRF tanks every Human weaponry in existance, even Nukes way above unless they customize it to be a high heat blast. It’s like saying shoot Superman with Kryptonite ammunition and missiles, of course superman would die.
Optimus can tank Quintessa’s beams who can kill, heal and practically destroy anything since she is one of the 12 creators of the TRF Multiverse who seconds the power of the AllSpark. Plus even tanked Unicron’s powers amped beams from Quintessa again. Unicron has 1/2 the power of the Multiverse and his brother is Primus who is the other 1/2.
Megatron tanked the full power of the AllSpark and his body remained intact, he only shut down since there was a power overload to his core. Plus it took a small piece of the AllSpark to bring him back to life which shows Megatron was practically in a coma and unconscious. Prime went Toe to Toe with Megatron and tore off one of Megs’s legs with just a backhand.

Thanos is bled by Iron Man with superfists. Iron man with a spinning leg traps Thanos’s hand with IG to the ground with ease. Thanos is stumbled badly by Quill throwing electromagnetic grenades. Drax with kitchen knives, kicks Thanos in the Knee and it hurt him (Thanos grunts). Spiderman holds Thanos’s fist back without the IG who has Helicopter level - parts of building level strength. Plus remember, the Stones boost Thanos individually when not glowing. When he fits Stones in his Gauntlet, see his nerves, they Glow in terms of Stone colour. That shows part of the Stones are in his blood once installed and individually boosts him when not used.
Thanos is weak without the IG, he can only predict his opponents moves before the fight happens which is his only advantage. Other than that he is really weak in terms of durability. The Hulk fight was with a boosted Thanos power stone. Even though the stone didn’t Glow, Thanos had a drive of the power stone in his blood to match Hulk’s strength. Without the IG, Hulk would have trashed him. Which is also why MCU never gave a Hulk vs Thanos rematch in Endgame since they avoided the feat inconsistency.

The IG only operates in the MCU so if Thanos fought Optimus in TRF Universe, the Power stone and a boost in durability is the only useful tools he would have. The time stone is for MCU, Reality stone is for MCU, Soul Stone is for MCU etc etc.

Plus don't bring up that New Avengers/TRF comic being a relation of Marvel to TRF Universe as the comic is canon now in IDW and Marvel mainstream Universes. The only one that remains is Marvel 80's TRF comic where Spidey shows up but that is Comic Marvel Megatron. Marvel 80's IG Thanos in that Universe would have control with his stones over Marvel's TRF.
It will be an interesting fight to see.
