Gonna disagree about round two. I think Mifune takes that one and has a significant speed advantage at that point in the story. Zoro undoubtedly has an advantage in raw strength, but he isn't surviving serious hits anyways (Very hard to knock him over, but still dependent on his internals like most mortal characters) and he isn't landing any.
Mifune's only fight of note happened against Hanzo the Salamander. Hanzo was famous for naming the Three Sanin and protecting his village from the surrounding countries using his small country as a war zone. He is also specifically noted as being exceptionally fast and a whole group of samurai fled when he showed up without even considering fighting him.
This is in a world where Kakashi cut a lightning bolt in half while still being much below kage level and this was well before the Naruto main series even started. By Shippuden Kakashi had grown much stronger and still wasn't quite a kage level fighter that could go toe to toe with people like Pain or Hanzo. Hanzo was admittedly defeated (during his life time) off panel by the Seven Paths of Pain, but they're comfortably kage as well. Plus them being meat puppets kinda makes it fair for most people to lose to them if they don't know what is going on.
Scaling this much is lame, but Mifune ain't done much. He just performed well against people who should also wreck Thriller Bark straw hat's shit. This wasn't too long after Skypeeonya and the Straw Hats still weren't shit in the speed department.
Round 3 Zoro wins pretty comfortably.
Mifune has confidence that he can keep Hanzo from ever getting a jutsu off (we see people do jutsus at extraordinary speeds in the melee all the time). Blocks a rush from Hanzo when he wasn't even nearby beforehand.
