Depends on starting distance.

Mid-high distance: KF wins, low difficulty. She can launch her ice blasts and keep Luke at range, or just freeze the ground around her so that he can't approach. Since she can do this to stop Barry approaching, she should be able to stop Luke approaching. From then, she wins based on him not having any long-range attacks and not having shown any resistance to extreme cold (that I can remember.)

Close range: KF again, but slightly higher difficulty. Here, it just depends on who strikes first. I'd go for KF (has reacted to Black Flash.) Also, she can do that thing where she turns into a frost could, and Luke isn't hurting her like that. That should be able to knock him back, she can rematerialise wherever she wants, and from there he loses for the same reasons she wins long range. Luke could one-shot, but I don't see him managing to hit first.

Also, prep time gives KF another advantage. Luke can't really do anything with it, but KF can (as she has all of Caitlin's knowledge, I think.) As a biologist, if she has info on Luke she may be able to find a way to penetrate his skin, like Claire did with the acid. She doesn't need that though. She wins anyway based on the reasons above.

MId to long range Killer frost wins Low diff or losses badly, she might have amazing powers but she is not exactly a good fighter even cisco can dodge her ice attacks so if luke doesnt act arrogant he could easily get away and beat her of course easier said than done.

Close range Luke cage wins midd-high diff a single punch would knock her out along with few of her ribs, of course him being very durable can give him a few extra seconds as to a regular human KF strieks with her ice so he should manage to get a hit before dying or completely immobile agaisnt her ice
