Karai: highly trained ninja, around the level of any of the individual Turtles (doesn't have their stats, but about as skilled)
She's fought Leonardo a few times, he's usually portrayed as being a tad better than her, but it's a solid rivalry (and he's the best of the Turtles). This fight, Leo was able to knock her down to get away, but we saw her shortly after, just fine (his goal was escape, not full victory)
She's also a bullet timer
She's an archer as well
Emiko: prodigious archer, better than Prime Earth Ollie (by statements at least, feat-wise, he has showings she hasn't replicated, so make of that what you will)
This was her first fight, where she amazed Ollie with her speed. Worth noting that he was injured from his fight with Komodo already
She's also a very capable h2h fighter
She's started carrying some trick arrows recently (as seen above)
Way I see it, Karai's better at close quarters, Emi's better at range, but they're both well rounded enough to be a solid match at any distance (and I think the starting distance is conducive to a match that will incorporate both range and cqc)
Not sure if I should give her the new sword for this fight or not, might give her too much of an advantage in close quarters against Emi (who is probably already at a bit of a disadvantage in cqc)
Not that Emi should be underestimated in cqc, however
