No, Vertigo was an imprint owned by DC comics and shared the same universe as the main DC universe (sorta), it's just that a lot of those comics published under the Vertigo line tended to be more darker or adult themed, and not so family friendly. Characters such as Swamp Thing and John Constantine both got their start and main runs in Vertigo. While they interacted from time to time with the main DC universe (Such as in the Search of Swamp Thing storyline), for the most part, it was pretty much it's own little world.

Anti Monitor is multiversal. They csn beat him only in specific "forms". In other words, it depends on the version of Spectre and Darkseid. Both vary from jobbers on roughly Superman-level hightier ( f.e. New52 Darkseid, Blackest Night Spectre) to neigh-omnpotent dieties (e.f. Soulfire Darkseid, unbound Spectre). The latter could defeat Anti-Monitor with their powers combined, since god-powers of both take source from Presence (unbound Spectre) and Source (Soulfire Darkseid) which are both multiversal as well. Two multiversals > One.
So if they get their god-boosts they win. If they don't, it's missmatch.
