Manji is a somewhat lazy samurai that is immortal and must kill a thousand men before he can be allowed to die
Rin is the heroine of Blade of the Immortal, along with the titular immortal swordsman Manji.
Member of the Ittō-ryū.
Former member of the Ittō-ryū, she is small in stature but the aggressive individual in the duo of herself and her giant companion, Isaku.
Ayame Burando is a doctor in the Blade of the Immortal series.
A naturally gifted sword fighter, Makie brought shame upon her family thus driving both she and her mother to prostitution. She fell in love with Anotsu Kagehisa, fought against Manji, but failed to kill him. Her skills were only flawed by her own troubles emotions.
Manji's younger sister. Her mind reverted to a child-like state after witnessing her brother (a criminal) kill her husband (a cop). As such, she was unable to even hate her brother for what he'd done and it became his duty to take care of her, failing to do so is what caused him to swear to kill one thousand evil men in return for the one hundred good men he had already killed.
An eight-hundred year old nun (that is also the meaning of her name) who's goal is to save all the lost souls in Japan. She is kept alive by the Bloodworms, which she also bestows to Manji, who doesn't really want them.
"Johnny" is a man who masquerades as a priest in order to assassinate his victims but when he tries his game on Manji, he ends up dead as he learns that shooting some people in the head doesn't kill them.
Leader of a ronin gang, he challenges Manji to a duel of blades after Manji kills his brother "Johnny". When Manji says he's put down the way of the sword, Shido becomes outraged and slaughters Manji's mentally retarded sister, Machi. In response, Manji swiftly kills Shido and his entire gang.
Prior to the death of Machi, O-yō appeared to be the hostess at the place where Manji and his sister were staying, although her role is never conclusively stated in the story.
Husband of Machi, he is a samurai and an officer of the law. However, he dies at the hands of a criminal he is pursuing, it is watching this event that causes Machi's mind to revert to a child-like state as the criminal who kills him is her brother Manji (Manji and Saitō didn't know of this bond at the time, although it was in the back of Manji's mind that he'd heard this man's name before).
