Royal Flush Gang I like. They'd make a great intro villain in a film--the Scarecrow to some other Ra's al Ghul. Granted, I have a soft spot for them from Booster Gold and Batman Beyond. As for Dr. Destiny, I'm shocked his worthiness is in question after his impressive use in Sandman.
But beside all that, if you'll look at the text you quoted, I did say "one of the first," not "the first." It's debatable of course, but there's no way "White Martians", who are basically just generic alien invaders and would be seen as knock-off "body snatchers", should've survived the first wave of cuts.
Darkseid and the White Martians going before Amazo is ridiculous.
I'm viewing it as a first movie villain. In doing so, no, Darkseid should not be the first villain. He should be the last one. Amazo on the other hand, would make more sense as a first movie villain. But I still wouldn't want him in it truthfully.
I love this top 5 :( IDK WHO TO CHOOSE!!!!! Thanagarians, I think, would look freaking amazing on screen. I think with the right visual effects, it would look absolutely amazing. Imagine them falling from the skies. Bloody awesome. Not sure if I want them to win though. Vandal Savage, as said by @DCFan1992, may be too much of a boring character for me... especially as the main villain. I can see him in a sequel though. Everyone else, I believe, would look good to start off the JLA movies.
